While previous studies have reported the generation of hepatic cells from fibroblasts, there is growing interest in understanding the mechanisms of hepatic reprogramming and its applications in regenerative medicine. <\/p>\n<p data-start=\"692\" data-end=\"914\">In this study, we demonstrated that <strong data-start=\"719\" data-end=\"755\">overexpression of Yamanaka factors<\/strong> can convert mouse fibroblasts, in the presence of an optimized hepatic culture medium, into cells that closely resemble hepatocytes under in vitro conditions.<\/p>\n<p data-start=\"916\" data-end=\"1199\">By examining the effects of \u06f2\u06f0 candidate transcription factors, we identified a <strong data-start=\"965\" data-end=\"1005\">combination of three factors\u2014Hnf\u06f4a, Cebpa, and Nr\u06f1i\u06f2<\/strong>\u2014that are capable of directing fibroblasts toward the hepatic lineage. When used alongside Yamanaka factors, these TFs significantly enhanced the efficiency of hepatic reprogramming. <\/p>\n<p data-start=\"1201\" data-end=\"1310\">The induced hepatocyte-like cells (iHep) generated using this method exhibited key hepatocyte-specific characteristics, including:<\/p>\n<ul data-start=\"1311\" data-end=\"1472\">\n<li data-start=\"1311\" data-end=\"1348\">\n<p data-start=\"1313\" data-end=\"1348\">Expression of liver-specific markers<\/p>\n<\/li>\n<li data-start=\"1349\" data-end=\"1366\">\n<p data-start=\"1351\" data-end=\"1366\">Glycogen storage<\/p>\n<\/li>\n<li data-start=\"1367\" data-end=\"1383\">\n<p data-start=\"1369\" data-end=\"1383\">Albumin secretion<\/p>\n<\/li>\n<li data-start=\"1384\" data-end=\"1398\">\n<p data-start=\"1386\" data-end=\"1398\">Urea production<\/p>\n<\/li>\n<li data-start=\"1399\" data-end=\"1436\">\n<p data-start=\"1401\" data-end=\"1436\">Low-density lipoprotein (LDL) uptake<\/p>\n<\/li>\n<li data-start=\"1437\" data-end=\"1472\">\n<p data-start=\"1439\" data-end=\"1472\">Indocyanine green (ICG) uptake<\/p>\n<\/li>\n<\/ul>\n<p data-start=\"1474\" data-end=\"1671\">The production of iHep cells through these novel approaches can provide new insights into the molecular nature of hepatocyte differentiation and pave the way for the <strong data-start=\"1606\" data-end=\"1639\">development of cell-based therapies<\/strong> for liver diseases.<\/p>\n<h2 data-start=\"1678\" data-end=\"1785\"><span class=\"ez-toc-section\" id=\"Keywords\"><\/span><strong data-start=\"1678\" data-end=\"1694\">Keywords:<\/strong><span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p data-start=\"1678\" data-end=\"1785\">Direct conversion;
